RESUMO

Testing for substance toxicity for living organisms is an important step in the development and adaptation of new drugs for various purposes. Analysis of the dependences between toxicological parameters of chemical substances for various test objects and physicochemical properties of these agents is a promising trend. Partition coefficient logP (logKow) was chosen as the key physicochemical parameter determining the toxicological parameters of the same substance for hydrobionts at different developmental stages. We found a correlation between decimal logarithm of the ratio of LCe50 for fish embryos to LCa50 for adult fish and logP. This dependence was found as a liner combination of equations obtained by drawing a trend line between experimental points and calculation of Pearson's correlation coefficient R.

RESUMO

Muscle extracts of some fish species, i.e. pike (Esox lucius), sterlet (Acipenser ruthenus), pink salmon (Oncorhynchus gorbuscha) and, to a lesser extent, perch (Perca fluviatilis) and Russian sturgeon, (Acipenser gueldenstaedtii) prevent the development of premature senescence of the human embryonic fibroblasts induced by the sublethal concentration of H2O2. Muscle extracts of other fish species tested, i.e. coho salmon (Oncorhynchus kisutch) and zander (Sander lucioperca), have not demonstrated this feature. Cell proliferation increased after the action of the senescence-inhibiting muscle extracts. Possible mechanisms of the action of nature biologically active compounds that interfere with the development of stress-induced cell senescence are discussed.

RESUMO

We revealed empirical dependences between common logarithm of a ratio of rat oral LD50 to LCa50 for adult fish and lgP for 50 different chemicals; and common logarithm of a ratio of the oral LD50 in rodents to LCe50 for fish embryos and lgP for 30 different chemicals. The dependences were obtained by constructing a trend line between experimental points and calculation of Pearson's R correlation coefficient as a measure of regression significance. These dependences can show the influence of substance lipophilicity on its toxicity for aquatic organisms comparing to mammals.

RESUMO

The researchers happen to face with suspensions in their chemical, biochemical and microbiological practice. The suspensions are the disperse systems with solid dispersed phase and liquid dispersion medium and with dispersed phase particle size > 100 nm (10-7 m). Quite often the necessity occurs to separate solid particles from liquid. To use for this purpose the precipitation in gravitation field can make the process to progress too long. In this respect an effective mode is the precipitation in the field of centrifugal forces--the centrifugation. The rotary speed of centrifuge rotor and centrifugation time can be set analytically using regularities of general dynamics and hydrodynamics. To this effect, should be written and transformed the equation of First and Second Newton Laws for suspension particle being in the field of centrifugal forces and forces of resistance of liquid and vessel wall. The force of liquid resistance depends on particle motion condition in liquid. To determine the regimen the Archimedes and Reynolds numerical dimensionless criteria are to be applied. The article demonstrates the results of these transformations as analytical inverse ratio dependence of centrifugation time from rotary speed. The calculation of series of "rate-time" data permits to choose the optimal data pair on the assumption of centrifuge capacity and practical reasonability. The results of calculations are validated by actual experimental data hence the physical mathematical apparatus can be considered as effective one. The setting progress depends both from parameter (Reynolds criterion) and data series calculation. So, the most convenient way to apply this operation is the programming approach. The article proposes to use the program Microsoft Excel and VBA programming language for this purpose. The possibility to download the file from Internet to use it for fast solution is proposed.

Neutrophylic phagocytosis is affected by a lot of factors, including phenylcarboxylic acids secreted by bacteria into blood to protect from phagocytosis. The paper describes the most likely mechanism of inhibiting action of these acids, which is associated with the formation of strong pi-complexes between the pi-bonds of the molecular benzene ring of the acids and pi-bonds of unsaturated fatty acid molecules of the neutrophilic membrane. This fact was used to calculate the minimum inhibitory concentration of an acid and to show that there may be saturation in inhibition from the concentration. The experiment wherein the acids were added to blood at varying concentrations has confirmed the inhibitory effect of the acids, as well as saturation. Data processing indicates the unifactor inhibition of phagocytosis and confirms the postulated exponential inhibition at acid concentrations of to about 10(-7) mole/l. There is saturation with further acid concentration increases and there is a further reduced phagocytosis due to the influence of pH at a concentration of more than 10(-5) mole/l.

BACKGROUND: Tracer studies are an important tool to obtain information about the processes involved in the immunological response. Colloidal gold is widely used as a tracer, but its small size of label can cause some difficulty during low-resolution analysis. To overcome this difficulty, we developed a new method to follow the route of tracer movement within lymph nodes. METHODS: We applied conventional X-ray analysis, X-ray fluorescence analysis (XFA) subtractional microscopy using synchrotron radiation (SR) beams, light microscopy, and ultrastructural analysis to study the distribution and quantity of colloidal gold coupled with albumin within rat parasternal lymph node 2, 4, 6, 8, and 10 h after intrapleural injection of the tracer. RESULTS: At all the time points XFA-SR revealed that tracer formed a circle with a maximum concentration in the node periphery. XFA-SR measured colloidal gold concentration in the nodes reached its maximum (0.5-0.75 weight %) in 6-8 h. Subtractional microscopy revealed superficially located groups of cells filled with colloidal gold tracer. Light microscopy and ultrastructural analysis confirmed that the tracer was concentrated in the reticular cells, situated in the sinuses of the node. Sinusoidal reticular cells concentrated tracer at much higher rates than sinusoidal macrophages. Four hours after injection, gold appeared in the lysosomes of the follicular reticular cells. At the same time point, evidence of antigen presentation was obtained. Antigen presentation proved to be an extremely rara event since only one ultrastructural incident was found in 150 analysed grids. CONCLUSIONS: SR is a valuable tool for the analysis of gold tracer passage within the living organism.
